Abstract
Stocks are more permanent methods of storing bacteria than plates. These stocks can be recovered by pouring them into small volumes of BG-11 and growing under low light levels. As the culture grows, it can be passaged into higher volumes and appropriate antibiotics can be added.
Steps
1. Do not add any antibiotics
Add your stock to 2mL
BG-11 medium in a test tube and grow it overnight.

Transfer this to a 50mL
flask and make up the culture to 10mL
in volume.
3.
Once it reaches a lime green colour (#32CD32), add a fresh medium to make the culture up to 50mL
culture in a 150mL
4.
Further, make this culture up to 100mL
in a 250mL
flask once it returns to lime green.
