Abstract

This protocol is for the preparation of 10% glycerol stocks of bacterial strains.

Steps

1.

From a streaked plate of the bacteria, pick one colony and inoculate into a test tube containing 10mL 1X LB

2.

Grow the culture till it reaches an OD ~ 0.6

3.

Into cryo-vials add 1.6mL culture and 0.4mL 50% autoclaved glycerol.

Note

This is for preparation of 10% glycerol stock. This can be changed according to the requirement.

4.

Flash freeze the tubes in liquid Nitrogen and store it at -80°C
